About Dig1

  • Dig1 is localized in the nucleus (judged by immunofluorescence). Tedford et al. 1997 PMID 9094309
  • Dig1-GFP is localized in the nucleus. Localization is unchanged by pheromone exposure. van Drogen et al. 2001 PMID 11781566
  • Dig1 contains a putative MAPK-binding sequence at residues 97-107. Metodiev et al. 2002 PMID 12029138
  • Dig1 interacts with Fus3, Kss1, and Ste12 as judged by yeast two-hybrid and co-immunoprecipitation. Tedford et al. 1997 PMID 9094309
    • GST-Dig2 precipitation experiments show that the Dig2/Ste12 interaction does not require the presence of either Fus3 or Kss1.
  • Dig1 and Dig2 are serve redundant functions for both invasive growth and mating response. Tedford et al. 1997 PMID 9094309
    • Deletion of either of Dig1 or Dig2 by themselves has minimal effect on invasive growth (judged by morphology) and on mating response (judged by growth arrest (BAR1 strain), and Fus1 gene expression).
    • Deletion of both Dig1 and Dig2 results in constitutive invasive growth.
    • Deletion of both Dig1 and Dig2 results in constitutive expression of the Fus1 gene (50-fold above WT cells), although exposure to pheromone still results in a modest increase in Fus1 gene expression.
  • Dig1 interacts with Kss1 via two-hybrid, co-immunoprecipitation, and in vitro. These interactions were largely unchanged by pheromone treatment. Cook et al. 1996 PMID 8918885
  • Dig1 that is co-immunoprecipitated with Kss1 can be phopshorylated in vitro by Kss1 when the cells are pre-treated with pheromone. Cook et al. 1996 PMID 8918885
  • Dig1 contains a putative MAPK docking site within residues 92-113, and Dig1(92-113) binds Fus3 and Kss1. Kusari et al. 2004 PMID 14734536
